A. Modified Method of in Utero Analysis of Sister Chromatid Exchange for Detection of Transplacental Mutagens

Abstract
Currently, several methods are being utilized to detect the genetic damage by transplacental mutagens measured by sister chromatid exchanges (SCEs). The techni-ques involve, for examples, 24 hour infusion of 5-bromodeoxyuridine (BUdR) via the mouse tail vein in one instance and BUdR tablet subcutaneous implant in another. The inherent difficulties of the time, quantity of BUdR, and become apparent of procedural difficulties involved.
Vogel & Bauknecht(1976) simplified the technique for in vivo analysis of SCEs in bone marrow cells of non-pregnant mice by repeat injecting BUdR intraperitoneally.
We modified this technique in order to apply it to pregnant mice. The SCEs in maternal bone marrow and fetal liver cells would be compared in order to predict the transplacental activity of the mutagenic agents in our case (cyclophosphamide, adria-mycin). This modified technique holds promise for the future as an easier, less time and BUdR consuming, and more accurate method than those previously utilized to detect genetic damage caused by transplacental mutagens.
